Na+,K+-ATPase and Cardiotonic Steroids in Models of Dopaminergic System Pathologies

چکیده
In recent years, enough evidence has accumulated to assert that cardiotonic steroids, Na+,K+-ATPase ligands, play an integral role in the physiological and pathophysiological processes body. However, little is known about function of these compounds central nervous system. Endogenous steroids are involved pathogenesis affective disorders, including depression bipolar disorder, which linked dopaminergic system dysfunction. Animal models have shown steroid ouabain induces mania-like behavior through dopamine-dependent intracellular signaling pathways. addition, mutations alpha subunit lead development neurological pathologies. Evidence from animal confirms consequences subunit. This review dedicated discussing pathologies—both supporting their involvement potential pathways along they may exert effects evaluated. Since there association between disorders accompanied by functional alterations such as Parkinson’s disease, we extend our discussion neurodegenerative diseases well.

Cardiotonic Steroids and Cardiac Fibrosis
It is well known and has been shown that patients who have chronic renal failure tend to develop and die of cardiac causes. These patients are known to develop a cardiomyopathy that is characterized by left ventricular hypertrophy and significant diastolic dysfunction. It has also been shown that the chronic renal failure condition is characterized by significant sodium pump inhibition due to i...
متن کاملCardiotonic Steroids as Modulators of Neuroinflammation
Cardiotonic steroids (CTS) are a class of specific ligands of the Na(+), K(+)- ATPase (NKA). NKA is a P-type ATPase that is ubiquitously expressed and although well known to be responsible for the maintenance of the cell electrochemical gradient through active transport, NKA can also act as a signal transducer in the presence of CTS. Inflammation, in addition to importantly driving organism def...
متن کاملStructural insights into the high affinity binding of cardiotonic steroids to the Na+,K+-ATPase.
The Na+,K+-ATPase belongs to the P-ATPase family, whose characteristic property is the formation of a phosphorylated intermediate. The enzyme is also a defined target for cardiotonic steroids which inhibit its functional activity and initiate intracellular signaling. Here we describe the 4.6 Å resolution crystal structure of the pig kidney Na+,K+-ATPase in its phosphorylated form stabilized by ...
متن کاملCardiotonic steroids-mediated Na+/K+-ATPase targeting could circumvent various chemoresistance pathways.
Many cancer patients fail to respond to chemotherapy because of the intrinsic resistance of their cancer to pro-apoptotic stimuli or the acquisition of the multidrug resistant phenotype during chronic treatment. Previous data from our groups and from others point to the sodium/potassium pump (the Na+/K+-ATPase, i.e., NaK) with its highly specific ligands (i.e., cardiotonic steroids) as a new ta...
متن کاملEpilepsy and dopaminergic system
Epilepsy is accompanied with a strong change in neuronal activity not only in excitatory (glutamatergic) and inhibitory (GABAergic) neurotransmission, but also in neuromodulatory agents. Dopaminergic system, as an important neuromodulatory system of the brain, has significant effect on neuronal excitability. In addition, this system undergoes many changes in epileptic brain. Understanding the e...
